• Note (General): type:text; アミド基は, 医薬品や化学繊維など様々な分野で重要な役割を果たしている。そのため, 有用なアミド基を作る方法が数多く開発されてきた。しかし, アミド基は作るのは簡単だが, 非常に安定なため他の物質への変換が難しい。本研究では, 簡単に作ることができるアミド基を他の有用な物質へ変換する求核付加反応を新たに開発した。この反応では, ジルコニウム試薬の特徴を利用して, 1回の反応で2つの試薬を一挙に導入できた。また, エステルなどの反応性の高い官能基が存在しても, これらを全く損なう事なく, アミド基のみ選択的に作用する優れた反応であった。また, 開発した反応を利用して, 生物活性アルカロイドの合成に成功した。 Amide functional groups are found in a wide variety of organic molecules including pharmaceuticals and functional materials. Therefore, efficient synthesis of amide bonds has been extensively studied. On the other hand, transformation of the generated amide groups is less explored than their construction. In this study, we developed direct nucleophilic addition to amide groups using unique properties of a zirconium reagent. The developed method enabled installation of two functional groups in a single reaction. The most conspicuous feature of our method is the high chemoselectivity. The reaction proceeded in the presence of a variety of sensitive functional groups such as esters.
