細胞遊走阻害剤UTKO1はホヤの脊索管形成を阻害したことから, UTKO1の標的タンパク質を同定することで脊索管形成分子機構を明らかにすることにした。まず, ビオチン標識化したUTKO1と脊索管形成期におけるホヤ抽出液を混合しアビジンビーズでプルダウンすることでUTKO1標的タンパク質を同定し, そしてその直接結合を大腸菌より精製した同タンパク質を用い確認した。さらに同タンパク質をモルフォリノアンチセンスオリゴヌクレオチドによりノックダウンしたところ, 脊索管形成がUTKO1同様に阻害されたことから, 実際に同タンパク質がホヤ脊索管形成に関与していることを示した。 In the present study, we aimed to elucidate the molecular mechanisms of Ciona notochord tubulogenesis by identifying the target protein of UTKO1, a cell migration inhibitor. Avidin beads pull down assay using biotin labeled UTKO1 and the lysate prepared by the larvae during notochord tubulogenesis identified the target protein of UTKO1, and we confirmed this direct interaction using the protein purified from E.Coli. Furthermore, knockdown of target protein using morpholino antisense oligonucleotide inhibited notochord tubulogenesis as seen in UTKO1 treatment, suggests the involvement of target in notochord tubulogenesis.
