Pin1はプロリンの構造をcis体からtrans体に変化させることでタンパクの立体構造を変化させるプロリン異性化酵素の一種である。種々のプロリン異性化酵素の中で, Pin1はリン酸化Ser/THr-Proのmotifに結合するという特徴を有し, リン酸化と構造変化の架け橋を行う。前立腺癌の臨床検体におけるPin1の強発現は強力な再発予測因子であった。また去勢抵抗性前立腺癌細胞株を使用し, in vitroにおけるPin1阻害の有用性を示した。この結果は去勢抵抗性前立腺癌の治療において, Pin1を治療標的とすることの有用性が示唆された。 The prolyl isomerase Pin1 regulates phosphorylated status of proteins by catalyzing the cis-trans isomerization of phosphorylated Ser/Thr and Pro motifs, and plays an important role in cancer progression. Strong expression of Pin1 in prostate cancer samples taken from prostatectomy was a potent predictive factor for prognosis. In vitro, inhibition of Pin1 showed significant anti-tumor efficacy for castration resistant prostate cancer cells. These results suggested Pin1 may serve as a promising therapeutic target for castration resistant prostate cancer.
