正常細胞は低酸素条件下で細胞死を引き起こすのに対して, 癌細胞は低酸素のような劣悪な環境でも生存できる細胞である。研究代表者らは, 低酸素による正常細胞の細胞死誘導過程でミトコンドリア内膜タンパク質Prohibitin が減少すること, ヒト大腸癌細胞においてProhibitinをノックダウンするとS期の割合が減少して細胞増殖が抑制されることを見いだした。さらに, Prohibitinとミトコンドリア内膜タンパク質Hint2との関連について解析を行った。 Cancer cells are more resistant to hypoxia than normal cells. We found that mitochondrial protein Prohibitin was decreased under hypoxic conditions, and that overexpression of Prohibitin inhibited cell death induced by hypoxia. Furthermore, Knockdown of Prohibitin in human colon cancer cells reduced S phase cell fraction and decreased cell proliferation. In addition, we analyzed functional cooperation of Prohibitin and mitochondrial protein Hint2 in colon cancer cells.
