腸内細菌叢と宿主粘膜免疫系に関連する近年の報告から, 主要な腸内細菌群の一つであるクロストリジウム目細菌群が, アレルギーや炎症性腸疾患などの抑制に重要な役割を担う制御性T細胞(Treg細胞)の分化・誘導を促すことが報告されたが, その分子機構は不明であった。そこでクロストリジウム目細菌群定着マウスに, 高繊維食または低繊維食を与えることで腸内環境に摂動を与え, その変化を統合オミクスにより解析した。その結果, 腸管内でクロストリジウム目細菌群が食物繊維の発酵代謝により産生する酪酸が, 大腸粘膜におけるナイーブT細胞からのTreg細胞への分化をエピジェネティックに誘導することを明らかにした。 Gut commensal microbes shape the mucosal immune system by regulating the differentiation and expansion of several types of T cell. Clostridia, a dominant class of commensal microbe, can induce colonic regulatory T (Treg) cells, which have a central role in the suppression of inflammatory and allergic responses. However, the molecular mechanisms by which commensal microbes induce colonic Treg cells have been unclear. We show that a large bowel microbial fermentation product, butyrate, induces the differentiation of colonic Treg cells in mice. An integrated omics approach identified gut microbiota-derived butyrate epigenetically induced the differentiation of Treg cells in vitro and in vivo, and ameliorated the development of colitis in mice. Our findings provide new insight into the mechanisms by which host– microbe interactions establish immunological homeostasis in the gut.
