本研究では, CD44vの発現やグルタチオン量が臨床において有用であるかを確認するために, セツキシマブ耐性口腔扁平上皮癌細胞を樹立し, 親株との性質の違いについて主に細胞内代謝に着目して検討した。その結果, セツキシマブ耐性細胞では, 抗酸化物質グルタチオンの産生を抑えるxCT阻害剤スルファサラジンに対する感受性が高まることが明らかとなった。このことから, スルファサラジンは, セツキシマブ耐性口腔扁平上皮癌細胞に対して高い効果を発揮すると考えられ, セツキシマブ不応症例に対する治療法の一つとなる可能性が示唆された。 The aim of the present study was to confirm the benefit of the appearance of CD44v and the amount of glutathione in clinical use, focusing on the intracellular metabolism, the nature of the parental strain was investigated by incubating cetuximab-resistant oral squamos carcinoma cells. According to the study, the cetuximab resistant cells sensitivity to xCT inhibitor sulfasalazine suppressing the production of antioxidants glutathione was increased. From this fact, sulfasalazine seems to be beneficial for cetuximab-resistant oral squamos carcinoma cells and might be considered as one of the treatments for cetuximab resistant cases.
