従来の接着培養法による歯髄幹細胞の分離では, 血球系や成熟細胞等が混入するため, 歯髄幹細胞の本質や再生能力を解明することに限界があると考えられる。本研究では, フロサイトメトリーにて歯髄組織より高純度の歯髄幹細胞を予期的に分離した。LNGFR Low+/THY-1 High+の分画より分離した細胞は高純度のコロニー形成細胞の集団であり, 高い増殖能と多分化能を有することが明らかになった。また, この細胞はマウス頭蓋骨欠損モデルにおいて骨再生を促進した。このことからLNGFR Low+/THY-1 High+細胞は骨再生治療において優れたソースとなる可能性が示唆された。 Human dental pulp stem cells (hDPSCs) isolated by plastic adherence, frequently contain contaminating hematopoietic cells and mature cells, which has limited our ability to investigate their basic biology and regenerative properties. In the present study, we prospectively isolated a pure population of hDPSCs from human dental pulp by flow cytometry. Prospectively isolated, dental pulp-derived LNGFR Low+/THY-1 High+ cells represent a highly enriched population of clonogenic cells ; notably, the isolated cells exhibited high proliferation and multi-lineage differentiation potential in vitro. The cells also promoted new bone formation to heal critical-size calvarial defects in vivo. These findings suggest that LNGFR Low+/THY-1 High+ dental pulp-derived cells provide an excellent source of material for bone regenerative strategies.
