チロシンキナーゼが機能する際の構造動態を明らかにすることは, 創薬や生命現象の解明に役立つ。本研究では, チロシンキナーゼに内在する構造平衡をNMR法により解析することで, チロシンキナーゼの活性を担う動的構造を明らかにすることを目的とした。c-Src, Fyn, c-Ablの大腸菌での発現・精製プロトコルを確立し, リン酸化状態を制御したサンプルを調製した。安定同位体標識サンプルのNMRスペクトル測定を行うことで, リン酸化状態や薬剤の結合に伴うNMRスペクトル変化の検出に成功した。このスペクトル変化がチロシンキナーゼの機能に関わる構造平衡を反映している可能性がある。 Tyrosine kinases are major targets for anticancer drug. To understand the dynamic regulation of tyrosine kinases, NMR analyses of the structural equilibrium were performed. In this study, E coli. expression system and purification protocol for c-Src, Fyn, and c-Abl were successfully established. NMR spectral changes upon phosphorylation of c-Src and upon binding to drugs were observed. NMR approach would be useful for understanding the dynamic processes in the regulation of tyrosine kinases.
