循環腫瘍細胞や播種腫瘍細胞と腫瘍幹細胞の類似性に着目した。肝芽腫, 横紋筋肉腫, 神経芽腫, ラブドイド腫瘍の細胞株や検体組織では, 腫瘍幹細胞マーカー候補のCD44発現は全細胞株で確認されたが, CD44v発現は低かった。CD13陽性肝芽腫細胞株(HuH6)をSCID-NODマウスに移植しても腫瘍形成はみられなかった。肝芽腫細胞株ではtranilastによるsphere形成能の低下が観察され, CDDPとtranistの併用群で腫瘍細胞障害の増強傾向が認められたが, 他の腫瘍株では増殖抑制効果が得られなかった。これより腫瘍幹細胞増殖制御はがん種により異なる機序が働くことが示唆された。 Similarity between cancer stem cell and circulating tumor cell (CTC)/disseminated tumor cell (DTC) was noticed in pediatric cancers. CD44, a stem cell marker candidate, was expressed on all cell lines and clinical tissue specimens including hepatoblastoma, neuroblastoma, rhabdomyosarcoma, and rhabdoid tumor, whereas CD44v expression was quite low. CD13 positive hepatoblastoma cell line failed to form tumors after administrated in SCID-NOD mice. Tranist downregulated the sphere formation in hepatoblastoma cell lines, and also downregulated the stem cell proliferation when administrated with CDDP, whereas did not affect the proliferation of other cell lines. Expression of stem cell candidates is modified in a different manner in each molecule even in a identical cell line. Proliferation of cancer stem cell seems to be controlled in a different mechanism in each cancer.
