マウス気管同所移植モデルにおいて, PPARγのリガンドであるPioglitazoneを用いて, 気管移植後の移植グラフトに対する拒絶反応を検討した。Pioglitazone投与群では対照群に比して, 移植気管グラフトへの成熟T細胞の浸潤は抑制されなかったが, 内腔の閉塞抑制, 制御性T細胞の浸潤増加, 炎症性サイトカイン産生の抑制が認められた。PioglitazoneがPPARγを介し, 炎症性サイトカインの産生を抑制し, 制御性T細胞を誘導することで, 気管移植後の拒絶反応を抑制していると考えられた。 The administration of pioglitazone, a PPAR gamma agonist, leads to a decrease in tracheal allograft luminal occlusions and the induction of regulatory T cell accumulation in the allograft in a mouse tracheal transplantation model. This indicates a protective role of pioglitazone against bronchiolitis obliterans. The potential mechanism of these effects includes a decrease in inflammatory cytokine production. However, the anti-inflammatory potential, and efficacy in the maintenance of transplant tolerance needs to be validated in the future.
