本研究では, 多様な免疫調節を担う脂質抗原受容体の機能解析を行うために, ナチュラルキラーT(NKT)細胞を制御する糖脂質抗原受容体CD1dに着目し, 受容体タンパク質に選択的共有結合形成可能な共有結合型CD1dリガンドの開発を目指した。報告者らは, 既知リガンドであるα-GalCerを基盤とした化合物デザインを行い, 共有結合形成能を有する反応性官能基を, アミド基を含むリンカーを介して, リガンドに導入した。合成化合物の一部は, α-GalCerと比較して, CD1dに対して高親和性を有するとともに, 高いサイトカイン誘導能を示した。 We focused on CD1d protein that can control the activation of natural killer T (NKT) cells through the presentation of self and foreign lipids, and planned to develop selective covalent ligands toward CD1d in order to enable the functional analysis and the modulation of CD1d. Based on the known CD1d ligand, α-GalCer, we designed novel covalent ligands containing an amide group as a linker. Some of the designed ligands showed the higher affinities to CD1d, and increased the cytokine production compared with α-GalCer.
