シナプス伝達の長期抑圧(LTD)現象は記憶・学習の基礎過程であり, シナプス後膜におけるAMPA受容体の減少によって起きる。LTDは, 1) AMPA受容体を足場分子から外す, 2) AMPA受容体をエンドサイトーシスする, の2つの過程からなる。これまで後者の過程を担う機構はよく分かっていなかった。また小脳LTDになぜデルタ2型グルタミン酸受容体(GluD2)が必要なのかも謎であった。今回の研究により, GluD2はAMPA受容体の足場タンパク質からの外れやすさを調節することが判明した。また神経活動に応じてエンドサイトーシスに必要な分子AP-2が集積しAMPA受容体と結合することが分かった。 It is believed that long-term depression (LTD) is mediated by AMPA receptor endocytosis at the postsynaptic site. During LTD, AMPA receptors are first liberated from their anchoring proteins at the postsynaptic site, diffuse to the perisynaptic site, where endocytosis takes place. However how AMPA receptors are selectively endocytosed remained largely unclear. In addition, it has been a mystery why the delta2 glutamate receptor (GluD2) is crucial for induction of cerebellar LTD. In this study, we found that GluD2 determined whether anchoring proteins were dissociated from AMPA receptors. In addition, we found how clathrin adaptor protein AP-2, an essential molecule for clathrin-dependent endocytosis, accumulated at the perisynaptic site and recognized the AMPA receptor complex during LTD.
