粘膜免疫系の正常な機能には, 免疫系細胞のみならず外部環境と接する上皮細胞による粘膜抗原の認識と選択的輸送が必要不可欠である。その中心的な役割を担うのは, パイエル板上皮層に存在するmicrofold(M)細胞である。本研究では粘膜面の宿主防御におけるM細胞の役割と微生物認識機構の解明を試みた。粘膜感染症に対するM細胞欠損の影響を調べた結果, M細胞欠損マウスでは粘膜感染に対する抵抗性が減弱することが判明した。自然免疫系には大きな異常は認められなかったが, 適応免疫系の異常が観察された。これらの結果から, M細胞は粘膜面における抗原特異的な免疫応答の発動に貢献していることが明らかとなった。 It remains unknown whether M-dependent antigen uptake also contributes to cellular immunity against mucosal pathogens. Here we investigated the contribution of M cells to Th17-dependent cellular immunity. Wild type (WT) and M-cell-null mice were orally infected with Citrobacter rodentium. After the infection, M-cell-null mice developed more severe infectious colitis compared to WT mice, as evidenced by exacerbation of body weight loss and fecal clinical scores. Importantly, colonic Th17 cells were decreased in M-cell-null mice compared to WT mice at the early phase of infection. In contrast, under the physiological conditions, there was no significant difference in the number of colonic Th17 cells between the two groups. Collectively, these results suggested that M-cell-dependent antigen uptake plays a key role in the protection against C. rodentium infection by facilitating antigen-specific Th17 induction.
