現時点までに計54名(15組のASD兄弟ペアと12組の健常兄弟ペア)を登録した。ASDとその非罹患同胞の2群間での脳灰白質体積の検討では, 両群で有意な差はみとめなかった。これは過去の報告と矛盾しない結果であった。さらに検定水準を下げた解析からは, 非罹患同胞と比較してASD群では扁桃体や被殻の体積が減少し, 中前頭回では増加していた。これらは過去のメタ解析より体積異常を指摘されている脳領域と一致していた。以上より, 非罹患同胞は, 臨床的にはASDと診断はされないが, ASDの病態に関連する脳領域において軽度の脳構造異常を有しており, この共通所見こそがASDの生物学的な所見である可能性が示唆された。 We have enrolled total 54 participants including 15 ASD and their unaffected sibling (US) pairs and 12 healthy control sibling pairs in this study. When we compared gray matter volume (GMV) between ASD and US, there was no significant GMV difference between groups in a voxel-based analysis. However, when decreasing the level of significance, we found ASD had smaller GMV in the bilateral amygdala and caudate and larger GMV in the middle frontal gyrus compared to UN. These brain regions have consistently been reported to be associated with ASD pathology from previous meta-analysis studies. Although UN is not clinically diagnosed as ASD, UN has mild structure abnormalities in the brain regions related to the pathology of ASD. We suggested that these common findings between ASD and UN may be a specific biological marker for ASD.
