本研究はアストログリアから産生されるL-セリンが脳虚血再灌流障害後に認められる神経の新生にどのように影響するかを明らかにすることを目的としていた。アストログリア特異的PHGDH欠損マウスに中大脳動脈梗塞(MCAO)を施したのちの梗塞領域の大きさにはflox/floxマウスと有意な差が認められなかった。初代培養神経細胞を用いたIn vitroの検討では, L-セリンがp70S6Kのリン酸化を促進することが確認された。p70S6Kはたんぱく質の合成を促進することで細胞増殖を刺激する因子であることから, L-セリンは神経新生を刺激する可能性が示された。 In this study, I aimed to clarify how L-serine , which is suplied by astroglia, affects on neurogenesis after ischemic reperfusion injury in brain of MCAO mouse model. The size of infarct region after MCAO in Astroglia specific knock out of phosphoglycerate dehydrogenase (PHGDH) was not significantly different from that in flox/flox mice. L-serine activated phosphorylation of p70S6K in primary cortical neurons in vitro. Because p70S6K is the protein that facilitates protein synthesis and cell proliferation, L-serine possibly stimulates neurogenesis.
