本研究では, オートファジーに注目し, 尿路上皮癌においてこれを制御することで新たな治療戦略を見出すことを目的とした。オートファジーを制御するコノフィリンを膀胱癌細胞株に暴露したところ, 抗腫瘍効果を認めた。さらに, 既存の抗癌剤との併用効果も認め, その機序を解明するためにHSP90という蛋白に着目したところ, 既存抗癌剤によって増加したHSP90が新規薬剤と併用することによって減少した。これにより, HSP90が新規薬剤の作用機序の一端を担っていることが推測される。さらにマイクロアレイ解析を行ったところGEP97という細胞表面の蛋白も新規薬剤の作用機序にかかわっていることが示唆された。 In this study, we focused on autophagy and explored the new strategy for urothelial carcinoma by inhibiting autophagy. By using Conophylline which regulate autophagy, we could confirm anti-tumor effect. In addition, Conophylline had the synergistic effect with cisplatin. To reveal this mechanism, we focused on HSP90. HSP90 induced by cisplatin was reduced by Conophylline, and it might be the key protein for this mechanism. Furthermore, microarray assay showed GEP97 was also associated with anti-tumor effect of Conophylline.
