くも膜下出血患者の血清中でコントロールと比較して発現が1/4以下に低下している49種の(hsa-miR-223等), 2倍以上に上昇している27種のマイクロRNA(hsa-miR-451a等)を同定した。モデルマウスでは, 頭蓋内圧上昇を5秒とすると, 海馬でモデル作成後1日で微小血管基底膜が脱落し, アストロサイトの足突起が基底膜から剥がれた。3日後には基底膜周辺のアストロサイトのGFAP染色性が低下し, 神経細胞自体の形態学的, 数的変化は認めずに神経機能が障害された。くも膜下出血後のNVU統合性の崩壊は微小血管周囲に始まり, 周辺アストロサイトの機能障害を介して, 神経機能障害に至る可能性が考えられた。 49 microRNAs those express in SAH patients' sera lower than 1/4 and other 27 microRNAs those expressed more than twice compared to those in control sera were identified. Degeneration of basal membrane, and peeling of endofeet of astrocyte from basal membrane was identified in hippocampus of mouse SAH model 1 day after procedure (duration of ICP elevation in the model was as long as 5 s). Three days after procedure, GFAP positive cell was decreased at the same site but no quantitative nor morphological change in neuron was observed. From the current result, we speculate degeneration of NVU starts abound microvasculature after SAH, and emerges to surrounding astrocyte that leads to neurological deterioration.
