人工多能性幹細胞(iPS細胞)より成熟肝細胞への分化誘導をより高い精度で行う手法として, 間葉系幹細胞を利用した分化誘導法と脱細胞化肝骨格を利用した立体的な細胞の分化誘導法の確立を模索した。間葉系幹細胞や肝細胞骨格を用いてiPS細胞から肝細胞への分化誘導を行い得られた細胞は, 成熟した肝細胞機能を有した細胞を得ることができたが, 成熟肝細胞に比較するとまで機能的には未熟であり, 臨床応用にはさらなる技術革新を要する。 To obtain more mature iPS induced hepatocyte like cells, we investigated a novel protocol by using mesenchymal stem cells and decellularized liver matrix. Mesenchymal stem cells and decellularized liver matrix could enhance the hepatic function of human iPSC-derived hepatocyte-like cells. However, these cells have immature function compared to primary hepatocyte. More technological innovations are needed to clinical application.
