神経変性疾患のメカニズム解明に向けて『神経細胞とグリア細胞の細胞間相互作用の重要性』が明らかとなりつつある。本研究では『生きたまま』の状況を反映した, 神経細胞-グリア細胞間の相互作用を解析可能な手法を開発した。特に, 異なる動物種から神経細胞及びグリア細胞を調整し, 種間の配列多様性を利用した, 次世代シーケンスとバイオインフォマティクスの活用により, 各細胞で生じているトランスクリプトームのスナップショットが可能となった。この手法を利用し, 疾患iPS細胞より調整したグリア細胞の異常を検出することが可能となった。 It is becoming apparent that cellular interaction between neuronal cells and glial cells contributes to pathogenesis of neurodegenerative disease. I have developed a system to analyze the transcriptome of cocultured neurons and glial cells without mechanical isolation. Using this system, I discovered unique features in neurodegenerative patient-derived astrocytes, which lead to novel therapeutic application.
