アクアポリン4(AQP4)は, 脳浮腫の病態に関与していることが明らかになってきた。そこで, 脳浮腫に対する分子標的創薬としてAQP4に注目し, 独自に開発した大規模スクリーニングから絞り込んだヒット化合物を評価した。その結果, 少なくとも1つの化合物がAQP4阻害作用を示した。また, ドッキング研究からその化合物は細胞外側から阻害をしている可能性が示唆された。次にAQP4のエンドサイトーシスのメカニズムを検討した。その結果, 細胞膜への輸送, AQP4タンパク質の安定化, 細胞膜からライソゾームへと輸送されるためのシグナルの存在が示唆された。 Aquaporin4 (AQP4) has been shown to be involved in the development of brain edema. We recently developed a high throughput-screening system for AQP4, as a target molecule for the treatment of brain edema. In this project, we identified at least one chemical compound that inhibits AQP4, by using a stopped flow apparatus with AQP4 proteoliposome. Computer based docking study revealed that this chemical may interact with the extracelluar domain of AQP4. Next, we examined molecular mechanisms of AQP4 endocytosis to understand the molecular regulation of AQP4. We have identified three signals for ; i) trafficking to the plasma membranes, ii) protein stability and iii) sorting to lysosomes.
