本検討の目的はヒト(日本人)でのβ細胞量の生理的, 病的変化を明らかにし, その調節機構を解明することである。我々は剖検膵標本および膵摘出術を施行された患者の膵組織を用いて, 日本人における膵α・β細胞量の肥満や糖尿病における変化を検討し, 2型糖尿病ではβ細胞量が46%減少していることを報告した(J Clin Endocrinol Metab 2016)。また動物では膵内脂肪蓄積がβ細胞障害を惹起することが示されているがヒトではその関与は少ないことを報告した(J Clin Endocrinol Metab 2017)。 Aim of the study was to explore physiological and pathological regulations in beta cell mass in humans (Japanese). We have examined effects of obesity and diabetes on alpha and beta cell mass in humans and found a reduction in beta cell mass by 46% in Japanese patients with type 2 diabetes (Inaishi J et al. J Clin Endocrinol Metab 2016). Deleterious effects of pancreatic fat accumulation on beta cell has been shown in animal studies, which is postulated as a cause of type 2 diabetes development. By examining human pancreas, we have shown that intra-pancreatic fat accumulation has little effect on beta cell mass and glucose intolerance (Murakami R et al. J Clin Endocrinol Metab 2017).
