背景 : 免疫電子顕微鏡法(以下, 免疫電顕)では, 通常の試料作製(2.5%グルタールアルデヒド(GA)及び1%四酸化オスミウム(OsO4)による二重固定エポキシ樹脂切片)では抗原のマスキングにより免疫電顕は困難となるため, 免疫電顕用のサンプル採取が難しい材料(皮膚や腎生検, 心筋生検などの臨床検体や希少な研究サンプルなど)については施行できなかった。しかし, 適切な抗原賦活化を用いれば通常のエポキシ樹脂包埋サンプルにおいても免疫電顕が可能なことが示され(山下, 顕微鏡, 2014)当研究室においても検討する必要性が生じた。方法 : 今回の検討ではマウス腎を用いて抗ヒトCD15マウスモノクローナル抗体(DAKO, ×100)を用いた包埋後染色法(2.5%GA＋1%OsO4二重固定エポキシ樹脂包埋, 金コロイド標識抗マウス二次抗体)について, ナトリウムエトキシドによる脱オスミウム・親水化処理に対する最適条件を検討した。前年度の結果より, ナトリウムエトキシドの希釈倍率を×10, ×100, ×1000とし室温30分反応させ, その後クエン酸緩衝液を用いた熱による抗原賦活化処理を行い, 一次抗体は4℃一晩, 二次抗体は室温60分反応後, 飽和酢酸ウランで5分染色し観察を行った。また, 今回の検討で得られた最適条件にて抗ヒトEMAマウスモノクローナル抗体(DAKO, ×100), 抗ヒトVimentinマウスモノクローナル抗体(DAKO, ×100)での反応も検討した。結果 : CD15陽性シグナルは腎近位尿細管微絨毛に見られ, ナトリウムエトキシドの希釈倍率が×1000で最も観察が容易であった。×10, ×100ではシグナルは見られるものの切片の損傷が激しく切片全体の観察が困難であった。また, 抗EMA抗体, 抗Vimentin抗体では陰性であった。まとめ : ナトリウムエトキシドによる処理は通常処理されたエポキシ樹脂切片に対して有効であった。しかし, 他の抗体での反応性を確認できなかったため, 他のサンプルやマウス腎にて反応する他の抗体を検索し直す必要があると考える。 Background : Immunoelectron microscopy using post-embedding method is difficult for usual epoxy resin embedding tissue (double fixation of glutaraldehyde and osmium tetroxide). We examined whether usual tissue sample is able to immunostain or not. Sample and Methods : Sample was mouse double fixed kidney (2.5% glutaraldehyde and 1% osmium tetroxide) that was embedded in epoxy resin. Ultrathin sections were processed by sodium ethoxide solution at a dilution ratio in the range of 10 times, 100 times, and 1000 times, and heat-induced antigen retrieval were performed for their sections. Anti-human CD15 antibody, anti-human EMA antibody and anti-human Vimentin antibody were used as primary antibodies, and secondary antibodies were labeled with colloidal gold (12nm). Results and Conclusion : Anti-CD15 antibody was positive, and the observation of sections processed by sodium ethoxide at 1000 times had best images. Anti-EMA antibody or anti-vimentin antibody was negative. The process by sodium ethoxide is useful for immunoelectron microscopy using post-embedding method.
