心筋細胞は胎児期に活発に増殖するのに対して, 生後は増殖能力を失ない終末分化細胞となる。成体心筋細胞の「若返り」を目指して, 成体に発現せず胎児期心筋に特異的に発現する転写因子をマイクロアレイで抽出した。その結果, 24の転写因子が胎児期心筋に特異的に発現し候補因子となることを確認した。これら遺伝子をレンチウイルスベクターで生後ラット培養心筋細胞に導入して, 心筋増殖を誘導する遺伝子をスクリーニングした。その結果, 新規転写因子Xにより心筋増殖が2倍に上昇することを見出した。さらに転写因子Xを発現するアデノ随伴ウイルスベクター(AAV-X)の作製に成功した。AAV-Xをマウス生体内に導入したところXの高発現をmRNAおよび蛋白レベルで確認できた。さらに1か月後のadultマウス心臓でBrdU assayを行ったところ, 興味深いことに成体心筋細胞でBrdU陽性の増殖する心筋細胞を確認できた。一方, 陰性対照では増殖心筋細胞は観察できなかった。以上より生体内においても新規転写因子Ｘにより成体心筋細胞の増殖・若返りの可能性が示唆された。今後さらに心筋梗塞モデルやTgマウスモデルでXによる心筋増殖の分子機構, 再生医療への応用を検討する予定である。 Heart muscle cells are terminally differentiated cells, and myocardial injury leads to heart failure. New therapy is highly demanded. We sought to determine the factor that can proliferate cardiomyocytes in vitro and in vivo. We screened 24 factors that express in embryonic cardiomyocytes. We found that factor X can induce cardiomyocyte proliferation in rat neonatal cardiomyocytes in vitro. We next developed AAV vectors expressing X constitutively. AAV-X injection into neonatal mouse revealed that cardiomyocytes could proliferate in vivo. We are now extending our research to adult myocytes and hearts after myocardial injury for heart regeneration therapy.
