複数臓器がんで共通してDNAメチル化亢進を示す遺伝子は、エピジェネティックに制御され細胞分化に寄与する遺伝子であり、これらが共通して発がんにおけるエピゲノム異常の標的になっていると考えられた。エピジェネティック機構による細胞の分化状態の破綻が、複数臓器で共通する発がんのメカニズムである可能性が示唆された。胃がんを他臓器がんから識別でき、原発不明がんの臓器診断に有用なDNAメチル化診断指標を同定した。同指標のDNAメチル化状態は非がん胃にも共通して観察されることから、胃の臓器特異的なDNAメチル化プロファイルががんでも保持されたものと考えた。 Genes encoding transcription factors involved in development and differentiation are commonly methylated in multiple organ cancers. Disruption of the differentiated state may be a common feature of DNA methylation alterations during carcinogenesis. Gastric carcinoma-specific DNA hypermethylation was identified based on methylome analysis of multiple organs. Our criteria may be useful for detecting the primary organ in cases of cancer of unknown primary site. Specific DNA hypermethylation was not observed only in gastric carcinoma but also in noncancerous gastric tissue. Specific DNA hypermethylation may not occur cancer-specific but organ-specific, and not be a functional event but a surrogate marker for gastric tissue.
