腎臓病があると、心血管イベントの発症率が有意に高いことが知られている。両者の関係は「心腎連関」という言葉で表現される。とくに慢性腎臓病においては血管石灰化病変の形成・進行が、心血管イベントの発症に大きく関与している。本研究では、慢性腎臓病における血管石灰化の分子機構の解明と治療標的分子の同定を目的として、培養細胞およびモデル動物を用いて検討を行った。慢性腎臓病においては、血管平滑筋におけるNF-kB活性が血管石灰化に重要な役割を果たしていることを明らかにした。 Vascular calcification is highly prevalent in patients with chronic kidney disease. The aim of the present study was to determine the role of pro-inflammatory nuclear factor-kB signaling for vascular calcification in chronic kidney disease. We found that the nuclear factor-kB signaling in vascular smooth muscle cells plays an important role in phosphate-induced vascular calcificaton in chronic kidney disease.
