骨格筋の質は筋機能のみならず、個体の寿命をも制御しうる報告が相次いでいる。しかしながら骨格筋の質とは一体なんであるかと言う本質的な問いに対する答えは全く明らかにされていない。研究代表者は骨格筋線維１本１本を取り囲んでいる基底膜を構成するECM成分こそが骨格筋の質の本質であると考えた。運動後の質の良い骨格筋と、徐神経や糖尿病などの質の悪い骨格筋とのECM遺伝子を網羅的に解析した結果、Lamb3遺伝子を見出した。これは筋再生過程で強く発現が誘導されAkt経路を介して骨格筋の再生・肥大を正に制御することを見出した。これらの結果は、骨格筋内のECMは筋機能を規定する重要な内部環境であることを示唆する。 Quality of skeletal muscle is determined not only muscle function but also life span of individual. However, the question of what is "quality of skeletal muscle" is not answered currently. From my previous study, I hypothesized component of extracellular matrix (ECM) in basal lamina covered a skeletal muscle fiber is the substance of "quality of skeletal muscle". I compared between "good skeletal muscle" such as trained muscle and "bad skeletal muscle" such as denervated or diabetic muscle using qPCR array and identified Lamb3 gene as a muscle quality control gene. Lamb3 gene is poorly expressed in normal skeletal muscle and strongly induced during muscle hypertrophy and muscle regeneration. Lamb3 activates Akt pathway, resulting in muscle regeneration and hypertrophy. These results suggested that muscle ECM is an important internal environment and their manipulation can control muscle quality.
