子宮脱細胞化骨格 (DUS) を用いた子宮再生の技術を臨床応用すべく、滋賀医科大学動物生命科学研究センターを霊長類実験の実施場所として選定・決定し、実施に必要な体制とチームを整えた。その基盤知見・技術を強固にするためラットを用いた研究も並行して行い、内膜欠損モデルにおいて、DUS移植により腺管構造を有する内膜を再構築することが出来た。しかし、その構築効率は必ずしも高くないため、DUSに子宮構成細胞に分化し得る細胞を予め搭載して再細胞化することにした。胚様体形成とWNT/CTNNB1経路の活性化を通じてヒトiPS細胞をプロゲステロン応答性の子宮内膜間質細胞へ分化誘導する方法を共同研究により確立した。 For clinical application of uterine bioengineering to patients with partial uterine defects, we selected the Research Center for Animal Life Science, Shiga University of Medical Research, as the place for primate experiments and prepared staff members and experimental setup. To obtain more solid data towards clinical application, we simultaneously performed rat experiments and found that placement of a decellularized uterine scaffold (DUS) led to the regeneration of endometrium with delineated glandular and luminal structures in rat endometrium defect models. The regeneration efficiency, however, was not as high as expected, so we decided to employ DUS loaded with uterine cells to enhance endometrial regeneration. As one of the most likely candidates as the loading cells, we generated progesterone-responsive endometrial stromal from human induced pluripotent stem cells and showed that WNT/CTNNB1 pathway plays a critical role in the differentiation and generation processes.
