本研究の目的は、いまだに病態の解明がなされていないFuchs角膜内皮変性症の患者に由来する疾患特異的ヒトiPS細胞を樹立し、これを当教室で開発した技術を用いてiPS細胞から角膜内皮細胞への分化誘導を行い、誘導した角膜内皮細胞を用いて小胞体ストレスを中心とした疾患進行のメカニズムを解析し、Fuchs角膜内皮変性症の病態を解明することである。また、得られたin vitro実験系を角膜内皮の小胞体ストレスのモデルとして利用することで、角膜内皮細胞の細胞死メカニズムの解析や、小胞体ストレス抑制の観点から角膜内皮細胞保護作用のある新規因子・薬剤の検索を行うことである。 We newly established iPS cells from Fuchs’ corneal endothelial dystrophy (FECD) for the investigation of this disease. Three different SNPs were found in the TCF4 gene from our FECD patients. We derived neural crest cells (NCC) from iPSCs and sorted integrin alpha 4 and p75 NTR- double positive cells. We compared the characteristics of C-NCC (control normal donor iPS derived NCC) and F-NCC (FECD patient iPS derived NCC). Real time PCR showed the upregulation of the ER stress marker (CHOP) in F-NCC. Cornea endothelial cells derived from F-NCC also showed the upregulation of CHOP, GRP78 and XBP1 indicating that these cells are ideal for screening of potential new drugs.
