本研究では、既存の金属ステントに超音波照射を行うことでフリーラジカルを発生させ、ステント内再狭窄を予防する治療法の開発を目指した。In vitroでは、超音波照射によるナイチノールの励起と、血管平滑筋細胞の増殖抑制効果を確認した。また、先行研究として、ミニブタ正常血管における金属ステントと生体吸収性ステントの長期的な開存性、血管壁反応、至適観察期間を検討した。結果、金属ステントでは経時的に狭窄率が増悪するのに対して、生体吸収性ステントでは留置後6週時点で最大狭窄となり、その後は徐々に狭窄が改善されることが確認された。 The aim of this study is to develop minimally invasive treatment for preventing in-stent restenosis of peripheral artery disease. An in vitro study, we confirmed excitation of nitinol and inhibitory effect of vascular smooth muscle cell proliferation by ultrasonic exposure. Furthermore, we conducted a study to comparison long-term vascular morphological changes between bioresorbable scaffolds and bare metal stents in porcine iliac artery. In this result, we found that the rate of stenosis of bare metal stents increased over time. In contrast, bioabsorbable scaffolds showed maximal stenosis at 6 weeks after placement and the stenosis gradually improved thereafter.
