癌は均一な細胞集団ではなく、一部に治療抵抗性で癌の再発転移に関係するがん幹細胞という細胞が存在すると考えられている。このがん幹細胞に対する治療を行うことができれば、治療成績が向上すると期待されている。一方で、COX-2という生体内で炎症が生じた際に誘導される酵素がある。我々は、咽頭癌の検体でCOX-2の発現が高い症例程、抗癌剤に対する感受性が低いことを確認した。またこのCOX-2を阻害することで癌細胞の幹細胞としての性質が抑えられ、抗癌剤に対する感受性が向上することを確認できた。 COX-2 is one of the two isoforms of cyclooxygenase, an enzyme that catalyzes the conversion of arachidonic acid to prostaglandins. COX-2 is considered to be associated with progression in various cancers, and its expression has been reported to have an impact on poor prognosis in head and neck squamous cell carcinomas (HNSCC). Furthermore, COX-2 expression has been reported to be associated with sensitivity of anticancer drugs. However, the precise mechanism of COX-2 on chemoresistance in HNSCC is not fully elucidated. The aim of the present study was to investigate the impact of COX-2 on cancer stem cell property, and to reveal its effect on chemoresistance. As a result, we found that COX-2 expression is associated with cancer stemness property, and inhibition of COX-2 has the possibility to improve chemosensitivity in HNSCCs. Our results indicate that COX-2 is an attractive target for the treatment of HNSCC.
