本研究では、患者の遺伝子変異を保持したiPSCを、3次元構造を持つ網膜組織オルガノイドに分化誘導し、病態解析を行った。これを用いて網膜色素変性の病態を解析し、メカニズムに介入する事で新規治療法の開発につなげるための解析を進めた。実際には、視細胞に分化誘導すると蛍光を発するレポーターを、患者由来iPS細胞に導入しておき、分化誘導して得られた視細胞の蛍光を指標とし、フローサイトメトリーにより回収した。そして、回収した網膜視細胞のサンプルを用いて、リアルタイムPCRなどを用いて病態に関連しうる分子の発現を明らかにした。主に小胞体ストレスに関する分子の動きに着目して解析し、治療開発向けた研究を行った。 The iPS cells generated from somatic cells of a patient with retinitis pigmentosa and a control were differentiated into three dimensional organoid culture to analyze the pathogenesis and to explore therapeutic approaches in the future. The organoid derived from the patient preserved the mutation. To analyze the photoreceptors, reporter gene which expresses fluorescence after differentiation was induced in the iPS cells, then the fluorescence positive cells were collected by flowcytometry to use the samples for real time PCR analyses. In particular, endoplasmic reticulum stress was analyzed for the future development of the therapeutic approach.
