オミックス解析による胃癌TCGA (The Cancer Genome Atlas) 分類は、多様性に富む胃癌間質をも分類し得る。線維芽細胞に着目しsubtype毎にがん微小環境の解明を試みた。我々のCAF (Cancer-associated fibroblast) 再構成モデルを4Subtypeで作製、発現遺伝子はそれぞれ異なっていた。4subtypeのCAF初代培養に遺伝子発現解析を行ったが、多様性のせいかCAF再構成モデルとの共通性は見出せず。TCGA胃癌組織RNAseqとの比較では、間質を豊富に含むGS (Genomically stable) 群で共通の遺伝子群を抽出し得た。 TCGA (The Cancer Genome Atlas) classification, which classifies gastric cancer into 4 subtypes by omics analysis, can also classify highly diverse characteristics of gastric cancer stroma. We focused on fibroblasts and attempted to elucidate the characteristics of the cancer microenvironment for each subtype. When the "CAF (Cancer-associated fibroblast) reconstruction model" that we have already used was made with 4 Subtypes, it was found that the gene expression status was different for each Subtype. Gene expression analysis was performed on 4 subtype CAF primary cultures, but due to the diversity, no commonality with the CAF reconstruction model was found. By comparing the gastric cancer tissue RNAseq of TCGA with the CAF remodeling model, a common gene group whose expression was increased in the GS (Genomically stable) group rich in stroma could be extracted. The possibility of elucidating the cancer microenvironment using this model was suggested in the GS group.
