本研究では、ALSを引き起こす原因遺伝子の特徴を持つアミノ酸配列を人工的に設計し、そのcDNAをマウスのROSA26遺伝子座へ導入することにより、人工ALSモデルマウスを作成、その分子機構を解析することを目的とした. 期間内にALS病態誘導人工遺伝子ノックインマウスラインを獲得できた。現在、計画繁殖を行っている. 一方、すでに我々が確立しているALSモデルマウス（核移行シグナル欠損FUS(△NLS-FUS）トランスジェニック）の解析から、脳脊髄で、多岐にわたるスプライシングの変化が初期より認められるとともに、シナプス可塑性に必要なSemaphorin 3Gの発現亢進していることを見出した. The aim of this study was to generate a novel ALS model, knocked-in mouse of artificially designing cDNA harboring with the characteristics of the ALS causative genes causing ALS and introducing into the ROSA26 locus to examined their cytotoxicity in vivo. We were able to obtain a line of ALS artificial gene knock-in mice. Currently, we are planning to breed them. On the other hand, from the analysis of our established ALS model mouse (△NLS-FUS transgenic), we found that a wide variety of splicing changes were observed in the brain and spinal cord and also that semaphorin 3G, which is required for synaptic plasticity, was upregulated in this mouse.
