本研究では、代表者らがマウスで特異的4因子を用いて初めて成功した2型肺胞上皮細胞への直接誘導（直接リプログラミング）に関して、ヒトSFTPCリポーター線維芽細胞を用いてヒト2型肺胞上皮細胞の直接誘導を試みた。選定した16の候補因子から転写因子の組み合わせによる条件検討により、最終的にリプログラミング因子として3因子を選定した。本誘導SFTPC陽性細胞に関して、詳細な遺伝子プロファイルを今後検討していく予定である。 Our group was the first to successfully induce type 2 alveolar epithelial cells in mice by direct reprogramming using specific four factors. In the present study, we attempted to directly induce human type 2 alveolar epithelial cells using human SFTPC reporter fibroblasts. From 16 candidate factors selected, 3 factors were finally selected as reprogramming factors by combination experiments of transcription factors. The detailed genetic profile of these induced SFTPC-positive cells will be investigated in the near future.
