食道癌に対する化学療法前検体を用いたRNAシーケンスの結果、化学療法不応群には、有意に間葉系細胞に特徴的な遺伝子の発現が亢進していることが示唆された。その検証を目的として、食道癌細胞株を用いて化学療法耐性株を樹立した。今後、新規治療開発を目指し、既存の薬剤ライブラリーを用いたスクリーニングを行っていく。腫瘍微小環境における細胞老化のメカニズムに関しては、腫瘍微小環境においてTGFβが強い細胞老化を惹起することを確認することができた。現在そのメカニズムの解明と、免疫担当細胞までを含めたSenescence細胞の役割について検討を続けている。 Using clinical samples of esophageal cancer from biopsy, we conducted RNA sequence and compared the gene expression profile between responder and non-responders. As results, the mesenchymal gene signatures were upregulated in non-responders. For validation, the drug resistant esophageal squamous cell carcinoma cell lines were developed. Aiming to identify the new drug target, the high throughput drug screening will be conducted. In terms of the role of cellular senescence in the drug resistance mechanism, TGFβ was found to induce senescence in the tumor microenvironment.
