本研究では、次世代イメージング「ライトシート顕微鏡・膨張顕微鏡法」を利用し腫瘍内不均一性を立体視する。我々は、この技術基盤として新規3次元イメージングパイプライン：DIIFCO法を、実際の臨床組織で実装に移した。膨張顕微鏡法も、光学顕微鏡の回折限界を乗り越える最新の研究手法である。腎癌組織マイクロアレイを利用して、我々は組織膨張下にDIIFCO法でin situ hybridizationを行うプロトコール開発を進めた。膨張顕微鏡法とDIIFCO法を組み合わせることで、ナノレゾリューションでRNA局在がわかるようになった。また、不均一なlncRNA発現も、細胞内局在が明らかになった。 In this study, we use next-generation cancer imaging "light-sheet microscopy/expansion microscopy" to stereoscopically visualize intratumoral inhomogeneity. We have implemented a new 3D imaging pipeline: DIIFCO method as the basis of this technology in actual clinical tissues. Expansion microscopy is also the latest research method to overcome the diffraction limit of optical microscopy. Utilizing a renal cell carcinoma tissue, we proceeded with the development of a protocol for in situ hybridization by the DIIFCO method under tissue swelling. By combining the expansion microscopy and DIIFCO method, RNA localization can be understood by nano-resolution. In addition, heterogeneous lncRNA expression was also revealed to be intracellularly localized in our methods.
