本研究ではヒト膀胱癌細胞株5637-ARを用いてアンドロゲン受容体阻害剤、放射線照射、DNA修復機構に関連するPARP阻害剤の併用療法の有効性について検討した。AR阻害剤単独とPARP阻害剤単独での放射線増強効果は得られたが併用では単剤と比べ殺細胞効果に有意差を認めなかった。その原因としてARシグナルとPARP-1との関連がPCR法で認められたため、AR阻害剤投与時にPARP-1の発現も低下させてしまうことが考えられた。 We examined the efficacy of combination therapy with anti-androgenic agent, radiotherapy and PARP inhibitor for bladder cancer cell line: 5637 with androgen receptor expression. 5637-AR cells revealed anti-androgenic agent and PARP inhibitor enhanced radiosensitivity. Combined administration of PARP inhibitor and anti-androgenic agent didn’t showed higher anti-tumor effect than single agent.
