口唇口蓋裂は本邦において600人に1人の割合で発生する頻度の高い先天異常であるが、その発生原因についての詳細は不明であり、予防的手段は存在しない。口蓋は胎生期に前頭鼻隆起と上顎隆起が癒合することによって発生し、その癒合不全が口唇口蓋裂につながると考えられる。本研究は、マウスの口蓋組織の正常発生の様子を組織学的に解析することによって、口蓋裂発症のメカニズムの一端を明らかにする目的で行われた。Fucciマウスを用いて間葉系組織の増殖が認められることを確認し、また口蓋組織のホールマウント染色標本の系を確立して血管を中心とする三次元組織の可視化に成功した。 Cleft lip and palate is a congenital anomaly with high frequency worldwide, accounting for about one in 600 birth in Japan. Its pathogenesis remain elusive and there is no prophylactic approach. Occurrence of cleft palate results from failure of adhesion of frontonasal process and maxillary processes. The purpose of this study was to elucidate the mechanism of physiological fusion of these tissues by histological analysis. Study using Fucci mice revealed huge proliferation of mesenchymal cells. Wholemount tissue model was also prepared to analyze 3-dimensional vessels.
