• Note (General): type:text; 天然糖脂質terpioside B（1）は、海綿Terpios sp. より単離・構造決定されたフコース含有スフィンゴ糖脂質である。1の構造的特徴として、天然に希少な２つの1,2-cis-α-L-フコフラノシド(α-Fucf)と、1,2-cis-α-D-ガラクトピラノシド(α-Galp)を含む五糖鎖がスフィンゴ脂質と結合している点が挙げられる。一方、1は、LPS刺激によるNO産生を顕著に抑制する興味深い生物活性を有しており、新たなNO産生阻害薬のシード化合物として大きく期待されている。そこで本研究では、1の初の全合成と構造活性相関研究、及びNO産生抑制機構の解明を目的としている。本研究の初年度である平成30年度は、1の全合成研究を行った。 まず、diethyl D-(−)-tartrate (2)を出発原料とし、脂質セグメントに相当する2-[(2R)-2-O-Bz-tetracosanoylamino]-3-O-Bn-4-O-PMB-17-methylphytosphingosine (3)を25工程で合成した。次に、3と、phenyl 2,3,4,6-tetra-O-Ac-1-thio-α-Glcpから10工程で合成した2-O-Bz-3,6-di-O-PMB-4-O-TBS-α-Glcp trichloroacetimidate (4) とのグリコシル化反応後、TBS基の脱保護により糖受容体5を合成した。次に、申請者らが開発したp-ニトロフェニルボロン酸触媒を用いた1,2-アンヒドロガラクトース6と1,6-アンヒドロ-β-ガラクトピラノシド (7)との位置及び1,2-cis-立体選択的グリコシル化反応を行うことで目的とする二糖鎖を位置及び立体選択的、かつ高収率で合成することに成功した。その後、6工程を経て、二糖供与体であるトリクロロアセトイミダート糖8を合成した。さらに、得られた8と5とのグリコシル化反応後、脱保護により、望む三糖鎖を有する糖脂質9までの合成を達成した。現在、糖脂質9に対するα-Fucfユニットの導入を検討中である。 Terpioside B (1), a natural glycolipid, was isolated from Terpios sp. 1 shows a potent inhibitory activity against LPS-induced NO release. Structurally, 1 has i) a unique pentasaccharide moiety; α-Fucf-(1,6)-α-Galp-(1,4)-[α-Fucf-(1,6)]-β-Galp-(1,4)-β-Glcp containing rare two 1,2-cis-α-Fucf units and 1,2-cis-α-Galp unit, and ii) a 17-methyl-C18-phytosphingosine moiety. Thus, this study was aimed to accomplish the first total synthesis of 1 and to reveal the structure-activity relationship of 1 regarding inhibitory activity against LPS-induced NO release. This year, we conducted synthetic studies toward the first total synthesis of 1. Firstly, the synthesis of 2-[(2R)-2-O-Bz-tetracosanoylamino]-3-O-Bn-4-O-PMB-17-methylphytosphingosine (3) was completed from diethyl D-(−)-tartrate (2) in 25 steps. Next, the glycosylation of 3 with 2-O-Bz-3,6-di-O-PMB-4-O-TBS-α-Glcp trichloroacetimidate (4), which was prepared from phenyl 2,3,4,6-tetra-O-Ac-1-thio-α-Glcp in 10 steps, followed by deprotection of TBS group gave glycosyl acceptor 5 in high yield with complete stereoselectivity. On the other hand, the glycosylation of 1,2-anhydrogalactose 6 and 1,6-anhydro-β-Galp (7) using a catalytic amount of p-nitrophenyl boronic acid proceeded smoothly to provide the desired α(1,4) galactoside with high regio- and stereoselectivities in high yield. And then, the α(1,4) galactoside was converted into trichloroacetimidate donor 8 in 6 steps. Finally, the synthesis of glycolipid possessing a trisaccharide structure 9 has been accomplished by the glycosylation of 8 and 5, followed by deprotection reaction. Introduction of α-Fucf units to 9 toward total synthesis of terpioside B (1) is now under investigation.
