• Note (General): type:text; 生物学的環境修復は、微生物を利用して水や土壌に含まれる有害な汚染物質を分解除去し、壊された自然環境の回復を図る技術である。この技術においては、汚染物質の分解除去に適する微生物を見出すことと、効率良く微生物を活動させるための担持材料を設計することが重要である。 本研究では、微生物の探索および担持材料の微構造設計を行った。多様な微生物を入手するために生物学的条件の異なる地域を採取地とした。すなわち、沖縄県南城市の島尻マージ地域、沖永良部島沖泊海岸の大型有孔虫化石密集層地域、宮城県仙台市青葉山粘土層地域を採取地とした。微生物群集は、ポリメラーゼ連鎖反応－変成濃度勾配ゲル電気泳動（Polymerase Chain Reaction Denaturing gradient gel electrophoresis：PCR-DGGE）により解析した。PCR-DGGEではDNAを増幅し電気泳動することによってバンドパターンが得られ、微生物の群集間の比較や材料に接着した微生物の評価に有効であった。また、各種材料の緻密な平滑面における微生物の接着性と代謝活性を比較した。対象とした材料は、ヒドロキシアパタイト（HA）、炭酸カルシウム、フッ化カルシウム、ケイ酸塩、カーボン、ポリ塩化ビニル、ポリウレタンである。微生物の接着性と代謝活性はHAとカーボンが良好であり、特にHAが優れていた。この結果から足場材料をHAに絞り、微生物の接着しやすい微構造設計に注力した。柱状粒子が絡み合い微細な気孔をもたらす微構造を目標とし、多孔質HAを水熱合成した。各種の微生物を含む懸濁液に多孔質HAを浸漬し、微生物の接着性と代謝活性を検討した。多孔質HAへの微生物の接着は平滑面よりも格段に良く、また代謝活性の観点からも、柱状粒子の絡み合いによって形成された気孔を有する微構造は、微生物の優良な足場になることが明らかとなった。今後は各種の微生物の性質に合わせて多孔質HAの形状と気孔構造を最適化する予定である。また、土壌への直接利用だけでなく、バイオリアクターへの応用も検討する予定である。 Bioremediation is an effective method to degrade and remove the toxic pollutants in water and soil by using microorganisms. Suitable microorganisms for bioremediation and highly designed supporting materials for microorganisms are required. In the present study, several microorganisms with different properties were hunt and investigated. They were obtained from the soil in Shimajiri Marge (Nanjo, Okinawa), from the sedimentary fossil bed of giant foraminifera (Okidomari, Okinoerabujima), and from the clay area of Aobayama (Sendai, Miyagi). A comparison of bacterial species was conducted by using polymerase chain reaction denaturing gradient gel electrophoresis (PCR-DGGE) on microorganisms. This analysis was effective in characterization of microorganisms and in evaluation of adhesion behavior of microorganisms on materials because of their band structure derived from DNA. The adhesion behavior and the total metabolic activity of microorganisms adhering onto the flat surface of several materials such as hydroxyapatite (HA), calcium carbonate, calcium fluoride, silicates, carbon, polyvinylchloride and polyurethane were compared. These results suggested that HA and carbon had good affinity for microorganisms, and the most excellent material was HA. For the application of HA as support materials for microorganisms, the design of microstructure is important. HA with porous microstructure by the entanglement of the rod-like particles were prepared through hydrothermal treatment. Porous HA were immersed in a suspension containing microorganisms obtained from the soil, and the adhesion of microorganisms on the HA was examined. The adhesion of microorganisms on the porous surface was excellent in comparison with the flat surface. The total metabolic activity of the microorganisms adhering onto the porous surface was higher than that of the flat surface without pores. More precise microstructure will be designed for HA and the applicability to the bioreactor of this material will also be considered in the near future.
