• Note (General): type:text; 多発性骨髄腫は形質細胞ががん化した難治性造血器腫瘍である. 近年, レナリドミドなどの免疫調節薬により予後の改善が認められている. しかしこれらでも抵抗性を示す多発性骨髄腫症例が存在しており新規薬剤の開発が急務である. 一方, 長鎖非コードRNA(lncRNA)は200塩基より大きなRNAで, タンパク質をコードしていないRNAである. 機能は, 転写調節, 発癌・癌進展などに関与し多岐にわたる. しかし, IMiDs抵抗性におけるlncRNA関連性についての研究はなく, 本研究ではlncRNAに焦点をあてIMiDs抵抗性を示す分子機序を解明する. レナリドミドはCRBNというE3ユビキチンリガーゼに結合することにより新たにIKZF1/3, CK1alphaといった基質が分解されることで, 細胞増殖抑制やアポトーシスを誘導されることが報告されている. しかし, 我々はそれらの基質が分解されても細胞死を誘導しない細胞株を見出している. レナリドミド感受性株及び耐性株についてDNA microarrayを用いて遺伝子発現量の差異をGene Set Enrichment Analysis (GSEA), 発現変動解析した. GSEAよりレナリドミド耐性細胞株群のみでエンリッチされたlncRNAのうち, 耐性株群で発現上昇しているものは1種類, 減少しているものは4種類であった. 発現変動解析では, レナリドミド耐性細胞株群で未処理時に上昇しているものは20種類, 処理下で上昇するものとして2種類同定した. また未処理において減少するものは25種類得られた. 一方, レナリドミド感受性細胞株群でレナリドミド刺激依存的において上昇しているものは４種類, 減少しているものは1種類であった.さらにregular PCRでも同様の発現変動が認められたlncRNAについて, レトロウイルスを用いてノックダウンしレナリドミド感受性に影響する検討している. 今後は, レナリドミド感受性に影響があったlncRNAについて分子機序を解明し, IMiDs抵抗性難治性多発性骨髄腫に対する新しい治療法の開発につながていく. Multiple myeloma (MM) is a hematological tumor that is characterized by malignant plasma cells. Recently, prognoses of the MM patients have been significantly improved due to treatment with immunomodulatory drugs (IMiDs) including lenalidomide. However, the prognosis of MM patients with cytogenetic abnormalities remains poor. Long non-cording RNAs (lncRNAs) are broadly defined as longer than 200 nucleotides that do not encode proteins. It has been described that lncRNA involved in cell fates including proliferation and apoptosis by regulating RNA processing and translation. However, it remains unclear how lncRNA regulates the resistance to IMiDs in MM. In this study, we focused on the mechanism of IMiDs resistance in MM targeting lncRNA. Lenalidomide binds to E3 ubiquitin ligase CRBN, resulting in degradation of IKZF1/3 and CK1alpha, followed by cell death and/or suppression of proliferation. We identified the lenalidomide-resistant cell line, even if those substrates were degraded in lenalidomide stimulation. We examined gene profiles in lenalidomide-sensitive and –resistant cell lines with or without lenalidomide using DNA microarray following Gene Set Enrichment Analysis (GSEA) and Differential Expressed Genes (DEG). In GSEA, one lncRNA and four lncRNAs are enriched in lenalidomide-untreated resistance MM cell lines group as up-regulated and down-regulated lncRNAs, respectively. In DEG, twenty lncRNAs and twenty-five lncRNAs were identified in lenalidomide-resistant MM cell lines group as up-regulated and down-regulated genes, respectively. I am attempting to show that identified lncRNAs regulate lenlalidomide-induced apoptosis using retroviral delivery of shRNA against human those lncRNAs. I need to reveal how identified lncRNAs involved in the sensitivity of MM cells to IMiDs in the future. The results will allow us to develop more effective drugs and therapy for IMiDs-resistant MM.
