• Note (General): type:text; 本研究は，従来の酵素を使用した剥離方法では細胞を損傷してしまう細胞剥離プロセスを改善するために，超音波振動を用いて細胞を損傷せずに剥離する方法を構築することを目的とした．以前の研究を参考に汎用性を高めることを目的として，医療現場でも用いられるポリスチレン製の培養ディッシュを用いた細胞の剥離を行なった．この方法では，培養ディッシュに超音波振動を付与できる加振装置をガラス基材と圧電素子を用いて製作して用いた．この装置と培養ディッシュの間をグリセロールで満たし，超音波振動を培養ディッシュに効率よく伝達した．本手法については，現在査読付きの論文雑誌に投稿し，リバイスを行なっている．リバイスの過程で，細胞を剥離するために必要な圧力やせん断力について，ハイスピードカメラによる波面の撮影と簡単なモデルを使って理論計算したところ，これまで音響流で剥離していたと思われた細胞の剥離が音響放射圧とスイープ振動により生じるスロッシングによるものであることが明らかとなった． さらに，本研究の細胞剥離方法を応用し，細胞シートを生成する手法も確立している．すなわち，レイヤー状に細胞が接着したシート状に剥離することができる．本手法では，シートが破れることなく剥離することを可能とするために，ランジュバン型振動子を用いて培養面を均一に揺らすことで安定して細胞シートを生成することができる．本結果は，超音波振動を用いた新たな細胞剥離方法として用いることが期待できる． また，振動面にくぼみを作り，振動子の振動モードを調節することで，ディッシュの中心を位相差顕微鏡により観察しながら均一に振動させることができるようになった．この結果は，今後超音波振動を用いて細胞の接着力を測定するための方法を確立するための予備実験となった．今後は本手法を用いて細胞の接着力を測定する予定である． The purpose of this study is to demonstrate a damage-free method of the cell detachment using ultrasonic vibration in order to improve the cell detachment process without enzymes. For the purpose of improving general versatility with reference to previous studies, cells were detached using a polystyrene culture dish for clinical use. In the method, a transducer capable of exposing ultrasonic vibration to a culture dish was made of a glass plate and a piezoelectric element. The space between the transducer and the culture dish was filled with glycerol and the ultrasonic vibration was efficiently transmitted to the culture dish. We submitted to the journal paper and are doing the revise. In the process of revision, the numerical calculations of the pressure and shear flow using simple models and wave motion observed by a high-speed camera shown that the acoustic radiation pressure and sloshing caused by the sweep vibration were dominant factors to cell detachment. Furthermore, we developed a method for generating a cell sheet by applying the cell detachment method in this study. In order to detach cell sheets without breaking, a cell sheet on a dish was shaken uniformly by using a Langevin transducer. This result is able to use as a new method of cell sheet detachment method. In addition, by making a hollow on the vibration surface and adjusting the vibration mode, the center of the dish can be uniformly vibrated with an observation by the phase contrast microscope. This result was a preliminary experiment to establish a method for measuring the adhesive strength of cells using ultrasonic vibration in the future. I am measuring the adhesion of cells using this method.
