• Note (General): type:text; T細胞移入療法は極めて有用ながん治療法であるが、活性化されたT細胞は持続的な刺激を受けて疲弊(Exhaustion)するため、免疫記憶を確立することが難しい。ステムセルメモリーT細胞(TSCM)は近年発見されたメモリーT細胞サブセットであり、ナイーブT細胞とセントラルメモリーT細胞の中間の性質をもつ。TSCMは生体内で強い増殖能力を示し、疲弊しにくいため、T細胞移入療法に最適なメモリーT細胞サブセットであると考えられている。 我々は活性化したヒトCD8+ T細胞を、Notchリガンドを発現するフィーダー細胞と共培養することでTSCM様細胞(iTSCM)を誘導する方法を確立した。iTSCMは担ガンマウス内で顕著に増殖し、他のT細胞サブセットに比べて強い抗腫瘍効果を示した (Kondo et al. Nat Commun. 2017)。またiTSCMにキメラ型抗原受容体(CAR)を遺伝子導入したCAR-iTSCMは、従来のCAR-Tに比べて強い抗腫瘍効果を示した。我々は次にiTSCMの代謝学的側面に着目した。活性化T細胞と比較して、iTSCMではミトコンドリアの新生や融合が観察され、ミトコンドリアの酸化的リン酸化が有意に亢進していた。またiTSCM誘導過程において、脂肪酸をミトコンドリアへ輸送するカルニチンおよびカルニチンと脂肪酸を結合させるカルニチンパルミトイル基転移酵素(CPT1a)の発現が亢進していた。さらにiTSCMでは細胞内中性脂肪の貯蓄が明らかになり、脂肪酸酸化依存的な酸化的リン酸化も認められた。その一方で、酸化的リン酸化を阻害するとiTSCMの誘導は阻害された。 酸化的リン酸化の亢進は抗腫瘍効果を増強することが知られており、iTSCMの強い抗腫瘍効果はこの特徴的な代謝メカニズムに依存している可能性が示唆された。またiTSCMをCAR-T療法へ応用することで、従来のCAR-T療法に比べてより抗腫瘍効果の高いCAR-T療法を開発できる可能性が示された。 Adoptive T cell therapy is a promising strategy for cancer immunotherapy. However, infused T cells frequently become functionally exhausted, and consequently providing poor prognosis after the transplantation into patients. Recently, a novel memory T cell subset, stem cell memory (TSCM) cells, is found in human and non-human primate as CD95+ naïve-like T cells. They show naïve T cell-like surface marker phenotypes such as CD62Lhi CD44lo CCR7+. Adoptive transfer of tumor-specific TSCM cells is expected to overcome this obstacle, since TSCM cells are close to naïve T cells, but are highly proliferative and long-lived and produce a large number of effector T cells in response to antigen stimulation. I previously reported that activated effector T cells can be converted into TSCM-like cells (iTSCM) by co-culturing with OP9 cells expressing a Notch-ligand, DLL1 (OP9-hDLL1) (Kondo et al. Nat Commun. 2017). In this study, I analyzed the metabolic features of iTSCM cells and the application of iTSCM cells for adoptive T cell therapy. I found that mitochondrial oxidative phosphorylation (OXPHOS) and biogenesis were enhanced, on the other hand glucose uptake was suppressed in human iTSCM cells. These human iTSCM cells also contained intracellular neutral lipid droplets and exhibited higher OXPHOS dependent on fatty acid catabolism. Tumor antigen-specific iTSCM cells with endogenous antitumor TCR or chimeric antigen receptors showed much stronger antitumor potentials than conventionally activated T cells in in vitro and humanized murine models. These data suggest that intracellular lipid metabolism governs strong antitumor ability of iTSCM cells. In addition, adoptive iTSCM cell therapy is a promising therapeutic strategy for cancer immunotherapy. In conclusion, I showed that Notch signaling induces TSCM cells and these cells have superior antitumor effects by using murine and human T cells. These results have already published in Nature Communications journal. Recently I am trying to clarify the mechanism of TSCM generation in term s of metabolism. I also challenge to develop a feeder-free method in order to apply to clinical trials.
