• Note (General): type:text; 神経細胞を結ぶシナプスは、神経伝達の場であるだけでなく、記憶・学習の形成を担う重要な部位である。近年、シナプス形成・機能の解析が精力的に行われ、その機構を支える１分子として細胞接着型Gタンパク質共役受容体 (Adhesion-GPCR, ADGR) が注目されている。ADGRは、種々の機能ドメインを有する細胞外領域と、GPCRに共通な７回膜貫通領域およびGタンパク質や他のシグナル分子と結合しうる細胞内領域からなるユニークな分子群であり、脳内に豊富に発現しているものの、その働きは不明な点が多い。我々は最近、小脳神経回路の要衝を担う登上線維 (CF)－小脳プルキンエ細胞 (PC) シナプス (以下、CFシナプス) において、PCに発現するBAI3が同シナプスの形成・機能を制御することを見出した (Kakegawa et al., Neuron 2015)。そこで本研究ではBAI3の生理的機能をより深く理解すべく解析を行った。 まず、BAI3のシナプス形成能を確かめた。成熟野生型PCにBAI3を過剰発現させ、CFシナプスの形成様式を観察すると、BAI3を強制発現させたPCにおいてCFシナプスの結合強度が有意に増加した。また、PCには、通常１本のCFのみが投射しているが、BAI3を強制発現した細胞では、近隣からの更なるCF入力が認められ、１つのPCに対して複数のCF入力が確認された。これら結果は、PCに発現させたBAI3はシナプス形成能を介して他のCF入力を誘導していることが示唆された。さらに、BAI3を介したCF入力は、BAI3の細胞内領域は必要とせず、細胞外領域のみが重要であり、特に、同領域への内在性リガンドC1qL1の結合が必須であることが各BAI3変異体の強制発現実験により分かった。以上の結果から、成熟小脳の神経回路においてもBAI3－C1qL1相互作用を介してダイナミックに改変されていることが示唆された。シナプス改変が生涯を通じて行われている事実はきわめて興味深く、今後、基礎・臨床両研究に有益な情報をもたらすものと期待される。 In the CNS, neurons transmit signals through the synapse, which is a crucial site for learning and memory. Recently, adhesion-type G protein-coupled receptors (Adhesion-GPCRs, ADGRs) are intensively studied as key molecules regulating synapse formation and functions. ADGR is a family consisting of a long extracellular N-terminal region with multifunctional domains, seven transmembrane domains and an intracellular C-terminal domain in which G proteins and several signal molecules bind. Despite of its unique structure, how AGDR works in vivo is still unclear. We previously demonstrated that BAI3, a member of ADGR family abundantly expressed at the synapses between cerebellar Purkinje cells and climbing fibers (CFs) from inferior olivary neurons (CF synapses), is crucial for the synapse development (Kakegawa et al., Neuron 2015). In this study, to clarify the functions of ADGR in the CNS, we focused on BAI3 in the mature cerebellum. First, to confirm the synaptogenic activity of BAI3 in adult, we overexpressed exogenous BAI3 into wild-type Purkinje cells in mature cerebellum. Interestingly, exogenous BAI3 strengthened CF synapse inputs and induced further innervations by neighboring CFs. These results indicate that BAI3 has an ability to induce synapse formation. Next, by introduction of several BAI3 mutants into wild-type Purkinje cell, we found that BAI3 brought a neighboring CF innervation through its extracellular N-terminal region, but not intracellular C-terminal region where several molecules bind to convey the intracellular signals. Surprisingly, the effect of BAI3 N-terminal region is dependent on the binding of its endogenous ligand C1qL1, which is released from CF terminals. Taken together, CF synapses are highly dynamic through BAI3-C1qL1 interaction even in adult. We hope that these results can be useful information for understanding of ADGR functions in vivo.
