• Note (General): type:text; 所属研究部門では血清応答因子Serum Response Factor(SRF)に結合するMegakaryoblastic Leukemia (MKL) 1の細胞内局在がアクチン動態によって制御され、前駆細胞から脂肪細胞への分化を誘導することを見出した。その現象を応用し、SRFを変異させることで核内MKL1とSRFの結合が物理的に減少し、下流の標的遺伝子の転写は抑制されるのかということ、SRF変異ヘテロ接合個体で腫瘍細胞を移植すると細胞増殖が抑制されるのかについて、メダカを用いたin vivo実験を行い、腫瘍抑制モデルとしての可能性を検討した。平成30年度にゲノム編集技術 CRISPR/Cas9を用いてメダカSRF遺伝子の改変を試みSRF変異4系統を樹立した。6塩基(2アミノ酸)欠失もしくは9塩基(3アミノ酸)欠失を持つメダカ系統は変異遺伝子ホモ接合個体であっても野生型との表現型の違いは確認できなかった。それに対し、2塩基付加によりフレームシフトを起こした系統、411塩基の欠失（DNA結合ドメインの1/3を持たない）系統では変異遺伝子ホモ接合個体は孵化できなかった。野生型個体と411 塩基欠失ヘテロ接合個体でそれぞれ20個体以上を用いて体長・体重を測定したところ、体長に有意な差は見られなかったが、体重には有意差が見られた(P=0.007)。SRFは多くの標的遺伝子を持つDNA結合転写因子であるが、SRF下流遺伝子としてgli1、ccn1などを選び、転写量の変化を解析した。その際、9塩基欠失ホモ接合個体、2塩基付加ヘテロ接合個体、411 塩基欠失ヘテロ接合個体及び孵化前胚ホモ接合個体を選び、対照群として野生型個体を用いた。しかし発現量は個体間の差異が大きく、遺伝子に変異を持つことによる発現には有意差がみられなかった。さらにSRF変異個体と野生型個体にマウス骨肉腫培養細胞の移植を行い、腫瘍増殖を比較した。ここでも個体間の差異が著しく、どちらの場合も1週間以降は生着せずに消失してしまう場合がほとんどであり、実験系として成り立たなかった。今後、生着する腫瘍を選択する必要がある。 We found that the intracellular localization of Megakaryoblastic Leukemia (MKL) 1, which binds to the Serum Response Factor (SRF), is regulated by actin dynamics and induces differentiation of precursor cells into adipocytes. Applying that phenomenon, we carried out in vivo experiments using medaka as a tumor suppression model. We tested whether the transcription of downstream target genes was suppressed by mutated SRF, and the proliferation of transplanting tumor cells into SRF mutant heterozygous individuals was suppressed. In 2018, we attempted to modify the medaka SRF gene using the genome editing technology CRISPR/ Cas9 and established four SRF mutant strains. In the medaka strain having a 6 base (2 amino acid) deletion or a 9 base (3 amino acid) deletion, no phenotypic difference from the wild type could be confirmed even in the mutant gene homozygous individuals. On the other hand, the homozygous mutant gene could not hatch in the line that caused a frame shift by adding 2 bases or the 411base deletion (having no 1/3 of the DNA binding domain). Body length and weight were measured using at least 20 wild-type individuals and 411 base deletion heterozygous individuals, respectively. No significant differences were found in body length, but significant differences(P=0.007) were found in body weight. We selected gli1 and ccn1 as SRF downstream genes and analyzed changes in the amount of transcript. At that time, 9-base deletion homozygous individuals, 2-base addition heterozygous individuals, 411-base deletion heterozygous individuals and pre-hatch embryo homozygous individuals were selected, and a wild-type individual was used as a control group. However, the expression level varied greatly between individuals, and there was no significant difference in expression due to mutations in the gene. Furthermore, transplantation of mouse osteosarcoma cultured cells was performed on SRF mutant individuals and wild-type individuals, and tumor growth was compared. Results showed that the difference of tumor growth between individuals was remarkable. In most cases, tumor cells disappeared from wild type fish within one week, and it did not work as an experimental system. In the future, it is necessary to select a tumor cell having high engraftment properties.
