• Note (General): type:text; 申請者は、以前より環状エノール炭酸エステルの脱炭酸反応によってナザロフ環化反応が進行し、多置換シクロペンテノン誘導体が立体選択的に構築できることを見出していた。本申請研究では、キラルな環状エノール炭酸エステルを基質とし、脱炭酸型ナザロフ環化反応によって基質に含まれる不斉点を起点にして、不斉転写反応を介して、光学活性シクロペンテノン誘導体の合成を目指した。 まず、キラル固定相による分取を経て調製した環状エノール炭酸エステルに対して、従来のルイス酸を用いる反応条件試したところ、多くのルイス酸で収率良く反応が起きるものの、不斉転写率は中程度と不十分なものであった。更なるルイス酸のスクリーニングにより、カウンターイオンを持たないトリスペンタフルオロボランを用いたところ、不斉転写率が飛躍的に向上することを見出した。また溶媒効果も顕著であり、低温にて低極性溶媒を用いる条件が効果的であることも分かった。最適化された反応条件は、幅広い基質に適用できる高い一般性を持っていることも分かったが、電子供与性基を有する基質では、不斉転写率が低下する問題点を残した。 上述の基質一般性における課題を克服すべく、反応機構について再考したところ、ルイス酸触媒を添加すること自体に問題があるのではないかと仮説を立て、無触媒条件にて加熱のみで反応が進行すれば、不斉転写率の低下を抑えられると考えた。そこで、基質溶液を触媒を加えずに加熱したところ、反応が進行し、不斉転写率が大きく向上することを見出した。特に、ナザロフ環化反応は熱的条件でも進行することは報告されていたが、高温条件が必要であったのに対して、本反応では比較的低温で進行したことは特長的であると言える。 We has previously reported that the Nazarov cyclization reaction proceeds by decarboxylation of the cyclic enol carbonate, and poly-substituted cyclopentenone derivative can be stereoselectively constructed. In this study, decarboxylative asymmetric Nazarov cyclization was developed through chirality transfer of a chiral cyclic enol carbonate. At first, the cyclic enol carbonate prepared by the preparative separation using a chiral stationary phase was treated with conventional Lewis acids, resulting in the moderate enantioselectivity. By further screening Lewis acids, it was found that tris(pentafluorophenyl)borane was best and the chirality transfer rate was dramatically improved. Also, the solvent effect was remarkable, and it was also discovered that the use of a mnon-polar solvent at low temperature was effective. The optimized reaction conditions was applicable to a wide range of substrates. However, in the case of a substrate having an electron-donating group, there remains a problem that the asymmetric transfer rate is lower. In order to overcome the above-mentioned issues regarding the generality of the substrate, the reaction mechanism was reconsidered, and it was hypothesized that there was a problem with the addition of the Lewis acid catalyst and the reaction proceeded only by heating under non-catalytic conditions. By doing so, it was thought that the decrease of the chirality transfer rate could be suppressed. Then, it was found that when the substrate solution was heated without an acid catalyst, the reaction proceeded and the chirality transfer rate was greatly improved.
