• Note (General): type:text; 目的 2017年に実施された2件の調査航海により，アラスカ沖と三重県沖から未記載属（新属新種）と思われるPhanodermatidae科の線虫類が採集された．簡易的な観察では両地点の線虫に形態的な差はなく同種と思われたが，両地点は5,000 kmも離れているため，移動力の低い線虫がそれほど広域に分布するかどうかは疑問であった．そこで本研究では，形態観察とDNA解析に基づいて，本種の分類学的位置を明確にして新種記載することと，両地点の個体群の系統関係を解明することを目的とした． 結果 微分干渉顕微鏡を用いてアラスカ9個体，三重10個体を詳細に観察したが，両地点の標本に明確な差異は見られず，同種の可能性が高いと判断された．本種は頭部表皮，歯状構造，生殖補助突起の形態でPhanodermella属に一致したが，1対ある雄性生殖器の一方が長くもう一方が短い点でPhanodermella属のいずれの既知種（両方とも短い）とも異なった． 一方，DNA実験にはアラスカ9個体，三重11個体を用いて，核18S遺伝子の約1600塩基とミトコンドリアCOI遺伝子の約260塩基を決定した．18Sは両地点の個体で最大1塩基の違いしかなく，同種または極めて近縁な種と判断された．COIは個体間で最大12%程度の変異を含むが，同地点の個体間よりも別地点の個体間の差異の方が明瞭に大きく，両地点間での自然交配は乏しいと考えられた．また，アラスカの方が遺伝的多様性が高く，祖先的な個体群の可能性が示唆されたが，解析に使用可能な個体が少なかったため確実ではない． 結論 以上の結果から，アラスカ沖と三重県沖のPhanodermatidae科線虫は同種とみなすのが妥当である．また，生殖器以外の形態ではPhanodermella属と区別できず，Phanodermella属のDNA情報がWEBデータベースにも存在せず既知種の配列と比較できないため，新属とみなすに十分な根拠があるとはいいがたく，Phanodermella属の新種とするのが無難と思われる．本研究の成果は2020年中に，DNAに基づく分布の議論を含めた新種記載論文として公表する見込みである． Introduction The specimens of undescribed free-living nematodes belonging to the family Phanodermatidae were collected from off Alaska and off Mie Prefecture in 2017. These nematodes seemed to be members of the same species based on the morphological similarity by the preliminary observation. However, it was doubtful because of the poor migration ability of nematodes to the great distance between habitats (about 5,000 km). In this study, morphological observation with a differential interference microscope and DNA analyses were carried out in order to reveal the taxonomic position and the phylogenetic relationship. Results Nine individuals from Alaska and ten from Mie were observed to conclude that they were in the same morphospecies. There were no clear differentiation between specimens from both sites. This species well agreed with the species of the genus Phanodermella in the structures of cephalic and pharyngeal capsules, and the presence of a precloacal supplement. However, it differed from them by having a long and a short male genital organs (vs. two short organs in Phanodermella species). DNAs were extracted from nine specimens from Alaska and eleven from Mie to decide the sequences of nuclear 18S (ca. 1,600 bp) and mitochondrial COI (ca. 260 bp) regions. In 18S, sequences from both populations completely agreed except only one base, so they considered to belong to the same or closely related species. In COI, sequences vary maximally 12% and suggested that currently restricted gene flow between populations. Also, it was suggested that Alaskan population was ancestral based on its higher diversity. Conclusion The phanodermatid nematodes from Alaska and Mie were considered to be in the same undescribed species. However, it was difficult to establish a new genus for this species, because it well agreed with Phanodermella species in the morphological characters except genital organs, and comparison of DNA sequences is impossible due to lack of data of Phanodermella is WEB databases. We will describe this species as a new species of Phanodermella with a discussion of distribution based on the DNA analyses in 2020.
