• Note (General): type:text; まず、C-結合型糖修飾を受ける疾患関連タンパク質を探索するにあたり、これまでC-結合型糖修飾が調べられた全タンパク質のコンセンサス配列周辺のアミノ酸配列解析を行った。その結果、コンセンサス配列周辺に分子量が大きいアミノ酸など特定のアミノ酸が存在するタンパク質ではC-結合型糖修飾が起こらないことが示唆された。そこで、これら知見を考慮した新規コンセンサス配列を仮定後、これら配列を有する疾患関連タンパク質をデータベースから絞り込み、変形性関節症の原因タンパク質であるADAMTS4やがんの転移およびアルツハイマー病の進行に関与すると考えられているCathepsin Bなど計8種類のタンパク質をC-結合型糖修飾における新規標的タンパク質とした。 続いて、これら新規標的タンパク質のC-結合型糖修飾を解析するためには、翻訳後修飾を受けた多量の標的タンパク質が必要となる。そこで、標的タンパク質遺伝子をクローニングしたのち、これら遺伝子をヒト培養細胞に遺伝子導入することで、それぞれの標的タンパク質の過剰発現ヒト培養細胞を樹立した。その後、標的タンパク質過剰発現ヒト培養細胞よりタンパク質を単離・精製し、質量分析により C-結合型糖修飾の有無を解析した。その結果、ADAMTS4では C-結合型糖修飾が予想されたアミノ酸全てで C-結合型糖修飾がされており、一方Cathepsin Bでは C-結合型糖修飾が予想されたアミノ酸ではC-結合型糖修飾はされていなかった。また、残り6種類の標的タンパク質については現在C-結合型糖修飾の有無を解析中である。 First, to explore novel C-mannosylated disease related proteins, we analyzed amino acids sequence around consensus sequence of the known C-mannosylated proteins. The results showed that proteins which include high molecular weight amino acids around consensus sequence were not C-mannosylated. Next, we presumed novel consensus sequence based on above results and extracted C-mannosylated candidate proteins, including the presumed consensus sequence, from protein data base. Consequently, we targeted total eight proteins such as ADAMTS4 and Cathepsin B as novel C-mannosylated proteins. Next, to analyze C-mannosylation of the targeted proteins, we tried gene cloning of the proteins and established the targeted proteins overexpression human cultured cells by the gene transfection. Additionally, we isolated and purified targeted proteins from the human cultured cells, and then C-mannosylation of the proteins were analyzed by LC-MS/MS. As a result, ADAMTS4 was C-mannosylated at all putative C-mannosylation sites, whereas Cathepsin B was not C-mannosylated. Currently, we are performing that C-mannosylation analysis of other targeted proteins.
