• Note (General): type:text; 昨年度にTH1細胞誘導に関与していると想定された遺伝子領域を欠損させたKlebsiella pneumoniaeを作成し、TH1誘導の強さが変化するかを検討した。その結果、欠損株では野生型と比較してTH1誘導は弱くなったものの有意な差はないことから、これら遺伝子領域以外にもTH1誘導に関与する遺伝子が存在することが示唆された。 そこで今年度は、様々なK. pneumoniaeを比較することによりTH1誘導に関与する遺伝子を再度絞り込むことにした。共同研究機関であるイスラエルのBiomX社から、クローン病・潰瘍性大腸炎患者の便から単離された１３株のK.pneumoniaeを譲受した。これら１３株のうち我々がクローン病単離した2H7株と同じmultilocus sequence typing (MLST)を持ち、ゲノム比較でもかなり近い株が6株あった。そこでまず、これら13株を２回の実験に分けそれぞれ無菌マウスに定着させ、１３株個々のTH1誘導の強さを検証した。その結果、強いTH1誘導を示す５株、中くらいのTH1誘導を示す株５株、弱いTH1誘導を示す株３株に分類できることがわかった。予想とは異なり、強いTH1誘導を示す2H7株と同じMLSTを持つ株は強い、中くらい、弱いTH1誘導それぞれに2株ずつ分類された。このことから、K. pneumoniaeのゲノム全体の相同性よりも特定の遺伝子がTH1誘導に関与している可能性が高いことが予想された。そこで、2H7株と同じMLSTを持つ6株を中心にゲノム配列を比較解析した。タンパク質をコードすると想定されるCDSのみで比較したところ、残念ながらTH1誘導と相関する遺伝子を見つけることができなかった。 以上のことからK. pneumoniaeのTH1誘導の強さはゲノム配列の相同性に相関しないこと、CDS配列の違いでは遺伝子を特定するのは難しいことが明らかになった。そこで今後はマウス腸内に存在しているK. pneumoniaeの遺伝子発現の違いを比較するため、単独定着マウスの便から細菌RNAを精製し、トランスクリプトーム解析を行う。同様にタンパク質発現を比較するため、便のプロテオーム解析を行う。 Last year, I created Klebsiella pneumoniae deficient in the gene region that was supposed to be involved in TH1 cell induction and examined whether the TH1 induction capacity changed. As a result, in the deficient strain, TH1 induction was weaker than that in the wild type, but there was no significant difference, suggesting that there are genes involved in TH1 induction in addition to these gene regions. Therefore, this year, we decided to narrow down the genes involved in TH1 induction again by comparing various K. pneumoniae. We obtained 13 strains of K. pneumoniae isolated from the stools of patients with Crohn's disease and ulcerative colitis from the collaboration research, BiomX of Israel. Of these 13 strains, 6 strains had the same multilocus sequence typing (MLST) as the 2H7 strain, and these 6 strains have quite similar genome sequences. I have assessed TH1 induction capacity of all 13 strains and it was found that 13 strains can be classified into three groups, 5 strains showing strong TH1 induction, 5 strains showing moderate TH1 induction, and 3 strains showing weak TH1 induction. Unexpectedly, 6 strains with the same MLST as 2H7 strain were classified into each of group. This data indicate that a specific gene is more likely to be involved in TH1 induction than homology of the entire genome of K. pneumoniae. Therefore, the genomic sequences of the 6 strains having the same MLST as the 2H7 strain were compared and analyzed. Unfortunately, we could not find a gene that correlates with TH1 induction when we compared only CDS, which is supposed to encode a protein. Thus, it was clarified that the TH1 induction capacity of K. pneumoniae does not correlate with the homology of the genomic sequence, and that it is difficult to identify the gene by the difference in the CDS sequence. Next year, in order to compare the difference in gene expression of K. pneumoniae in the mouse intestine, bacterial RNA will be purified from the feces of monocolonized mice and transcriptomic analysis will be performed. Similarly, stool proteomic analysis will be performed to compare protein expression.
