本検討はまだ研究の途中であり、学会・論文発表を行なっていないため、詳細な結果説明は行わない。本年度は、予定通りMycobacterium ulceransを標的に、複数のL A M P 、R P Aプライマーセットを作製した。１０倍希釈系列のD N A水溶液を使用して検出感度を測定し、最も検出感度が高いプライマーセットを明らかにした。特異度判定のために、３０種類の抗酸菌D N A、１５種類の細菌D N Aを使用し、特異度に問題がないことを明らかにした。本年度は、予定よりも早く検討が進んだため、LAMP、R P AとD N Aクロマトグラフィを組み合わせる方法についても検討を開始した。詳細は記載できないが、L A M P、R P Aクロマトグラフィについては、conventionalなL A M P、R P A法と検出感度は同等であった。共同研究者から、臨床検体由来のD N Aを分与してもらい、評価を行ったが、conventional、DNAクロマトグラフィ法ともに良好に検出が可能であった。次年度は、ガーナのフィールドで本法を応用してみて、検査実施に際して生じる様々な問題点を明らかにし、改善を進めていく予定である。 We designed multiple LAMP and RPA primer sets targeting Mycobacterium ulcerans using PrimerEplorerV (EIKEN Chem, Japan). We evaluated the detection sensitivity using a 10-fold dilution series of DNA solution, and identified the best primer set of LAMP and RPA. We also evaluated the specificity of the primer set using several DNA from mycobacterium and bacteria. In this fiscal year, the study proceeded earlier, therefore we developed detection method of LAMP, RPA combined with DNA chromatography. We found that the detection sensitivity of the LAMP and RPA chromatography was equivalent to that of the conventional methods. We will apply this method for patients in Ghana, and improve this method.
